If you are using it as lumber, I'd wait until it has dried. If you still green wood, it may twist/warp as it dries and your hole will no longer be straight. If that doesn't matter then I don't think it matters when you drill.

Either way, there are a few options. I'd use a ship auger....a regular drill bit may do it, but probably isn't cheaper. Krait makes speed bore bits... probably long enough: https://www.lowes.com/pd/IRWIN-SPEE...UuFTDgX02SfkmNohAqhoCsG0QAvD_BwE&gclsrc=aw.ds

A forstner bit will make a cleaner hole, but 6" is a lot for that.
